Abstract:

Background: The gut–brain axis has emerged as a major focus in contemporary medicine, highlighting the bidirectional communication between the gastrointestinal tract, enteric nervous system, microbiota, immune signaling, endocrine pathways, and the central nervous system. Although these mechanisms have gained scientific recognition only in recent decades, classical Ayurvedic literature describes the gastrointestinal tract as the foundation of digestion, transformation, nourishment, and systemic regulation. Revisiting these classical concepts through a scientific lens may provide novel insights for integrative lifestyle medicine.
Objective: To propose a conceptual hypothesis correlating selected Ayurvedic principles—particularly Pur??adhar? Kal?, Ap?na V?ta, and Agni—with the modern gut–brain axis and to explore their potential relevance in understanding intestinal physiology, systemic homeostasis, and integrative therapeutic strategies.
Content: This paper presents three interconnected conceptual hypotheses. The first hypothesis explores the physiological significance of the large intestine beyond fecal storage. While modern physiology attributes most nutrient absorption to the small intestine, the colon is now recognized for absorbing water, electrolytes, and microbiota-derived metabolites such as short-chain fatty acids. Accordingly, Pur??adhar? Kal? is conceptually interpreted as a broader functional interface involved in absorption, regulation, and maintenance of homeostasis rather than solely in fecal retention. The second hypothesis revisits the classical Ayurvedic emphasis on Sneha-based (lipid-based) formulations. Rather than suggesting that Ayurveda explicitly describes modern anatomical structures such as the ileocecal valve or other one-way valve mechanisms, this hypothesis proposes that lipid-based pharmaceutical preparations may have been designed to improve gastrointestinal residence time, facilitate tissue penetration, enhance bioavailability, and optimize systemic therapeutic delivery. This concept demonstrates a potential functional parallel with contemporary lipid-based drug delivery systems.
The third hypothesis proposes a conceptual relationship between Pur??adhar? Kal? and the modern gut–brain axis. Although classical Ayurvedic texts do not explicitly describe a direct anatomical connection between the intestine and the brain through Pur??adhar? Kal?, its close association with Ap?na V?ta, digestive regulation, and systemic homeostasis suggests a plausible functional analogy with contemporary neurogastroenterology. Disturbances in intestinal physiology may influence neurological, emotional, and cognitive functions through neural, immune, endocrine, and microbial pathways, providing a theoretical bridge between Ayurvedic concepts and modern biomedical science. In the digital era, increasing psychological stress, sedentary lifestyles, processed food consumption, circadian rhythm disruption, and prolonged screen exposure are contributing to impaired gut health and altered gut–brain communication. Integrating Ayurvedic principles with current gut–brain axis research may therefore offer innovative perspectives for preventive, personalized, and lifestyle-based healthcare.
Conclusion: The proposed hypotheses do not claim historical equivalence between Ayurvedic concepts and modern biomedical discoveries. Instead, they present a scientifically cautious conceptual framework intended to stimulate interdisciplinary dialogue, generate testable hypotheses, and encourage future translational research. Re-examining classical Ayurvedic wisdom alongside emerging evidence on the gut–brain axis may contribute to the development of evidence-informed integrative lifestyle medicine.
