Title : Evidence of immunomodulatory activity following administration of homeopathic remedy in healthy volunteers: A pilot controlled study
Abstract:
Background: Experimental evidence supporting the biological activity of homeopathic high dilutions remains limited, particularly in healthy individuals. Characterizing early immune responses induced by homeopathic medicines may contribute to understanding their potential mechanisms of action. Atropa belladonna is a well-established homeopathic medicine traditionally prescribed in acute inflammatory and febrile conditions, making it a suitable model for investigating whether administration to healthy subjects is associated with measurable changes in immune mediators. Inflammatory cytokines represent sensitive biomarkers for detecting subtle immune activation and may provide objective evidence of biological responses following exposure to highly diluted preparations.
Objective: To investigate whether repeated administration of homeopathic Atropa belladonna 200C is associated with changes in circulating inflammatory cytokine concentrations in healthy volunteers.
Materials and Methods: A pilot controlled study was conducted in six healthy volunteers allocated to an experimental group (n = 3) and a control group (n = 3). Peripheral blood samples were collected at baseline and 24 hours after intervention. Participants in the experimental group received Atropa belladonna 200C diluted in water, whereas controls received an indistinguishable hydroalcoholic solution. Both groups administered one dose every two hours during a ten-hour period. Vital signs—including body temperature, blood pressure, heart rate, respiratory rate, and oxygen saturation—were monitored throughout the intervention. Plasma concentrations of IL-1β, IFN-α2, IFN-γ, TNF-α, MCP-1, IL-6, IL-8, IL-10, IL-12p70, IL-17A, IL-18, IL-23, and IL-33, together with total immunoglobulins, were quantified using multiplex flow cytometry. Statistical analyses compared post-intervention values with baseline and between groups, considering p < 0.05 statistically significant.
Results: No significant differences were observed in vital signs or immunoglobulin concentrations following intervention. In contrast, participants receiving Atropa belladonna 200C demonstrated significant increases in selected inflammatory cytokines compared with baseline. Mean percentage increases were observed for IL-6 (93.87%), IL-8 (85.04%), IL-12p70 (84.57%), IL-23 (70.16%), IL-33 (55.19%), and IL-18 (53.15%), with statistically significant differences (p < 0.05). No significant changes were detected for IL-1β, IFN-α2, IFN-γ, TNF-α, MCP-1, IL-10, or IL-17A. These immunological changes occurred in the absence of clinically detectable inflammatory manifestations.
Conclusions: Repeated administration of homeopathic Atropa belladonna 200C was associated with selective modulation of inflammatory cytokine production in healthy volunteers while preserving normal clinical parameters. Although preliminary and limited by the small sample size, these findings provide proof-of-concept evidence that a highly diluted homeopathic preparation may be associated with measurable immunological responses under controlled experimental conditions. Confirmation through adequately powered, randomized, double-blind studies is warranted to determine the reproducibility, magnitude, and biological significance of these observations.
